Definitely check out a Broadway show. As they say, there's nothing like a show on Broadway...but make sure you stop by TKTS for discounted tickets first. Shows are magnificent but the price can be, too.
If you're looking for cheaper entertainment (or it's your first time visiting NYC) go for a FREE ride on the Staten Island Ferry. It goes right past the Statue of Liberty & Ellis Island while giving you a great view of Lower Manhattan. (For $12 you can actually visit the Statue of Liberty & Ellis Island).
For more great views of the City, go for a walk across the Brooklyn Bridge (free), and grab a bite at Grimaldi's (fantastic pizza on the Brooklyn side) or walk the other way and head to Lombardi's (the oldest pizzeria in Manhattan on Spring & Mott) Stop by Ferrara's Bakery in Little Italy for dessert & thank me later ;)
If the weather is nice, just pick a neighborhood and go for a walk. You really get to know the place better that way. Or if it's bad outside, head to any of the world famous museums. My favorites are the Metropolitan Museum of Art (pay-what-you-wish) and the Museum of Natural History. After exploring those for a bit, go for a walk through Central Park and visit the tribute to John Lennon at Strawberry Fields (in Central Park around 72nd St).
