It depends on what do you want to do. There are quite a few activities that you can do.
1) You may visit Sentosa Island (http://www.sentosa.com.sg/en/). You may also go to Universal Studio in Sentosa.
2) If you like the nature & wildlife, you may visit Pulau Ubin. You can do cycling and visit Chek Jawa. (http://www.wildsingapore.com/ubin/index.html)
3) You can also do the Duck tour (http://www.ducktours.com.sg/)
4) You may go to the Zoo or Night Safari (www.zoo.com.sg/).

There is a list of must see/do things for 1st timers to Singapore on NileGuide here:
The Singapore RIver Cruises or the Singapore Flyer is a good way to do some sightseeing in a short space of time. Also a wander around Chinatown is always nice for the atmosphere.

It depends on wat you want to do. How about going to east coast n enjoy the sea breeze while having a glass of beer and see the locals having fun at the beach. Or you can visit the zoo. I guess others had already told you bout our best attraction, so I wont go that way. Singapore is a great place to hang out and chill. There is alot of things that you may interest you. Its good fun when just walk around and found some place that suits you. That would be a experiennce.

There are so many places to visit here in Singapore... from the Birds Paradise, Singapore Flyer, Night Safari, Universal Studios Singapore and many more... in fact every part of it, whether the city or its country side would be a good scenery to cherish and appreciate... just a walk at the park or see the lights on a city night will surely leave you a feeling of happiness.

Start with a walk along the Henderson Bridges in the morning to whet your appetite and walk on down to Seah Im Hawker Centre where you can find favourite local delights (try the chicken rice!). Bask in the atmosphere along the Sentosa Boardwalk towards Sentosa and have a fun day exploring the biggest entertainment offshore island in Singapore. It will take about a full day. Head on back to Singapore and take the train down to Little India for a full cultural exposure to one of the heritage sites in Singapore. End your day with good food and drinks anywhere you fancy in niche cafes along Arab St. Wake up the next day and head on down to any of the Sunday Flea Markets available (Zouk, Raffles Place) or catch the ceremonial procession of guards changing at the President's Istana in Dhoby Ghaut (first Sunday of every month I think - gotta check it up). Soak it up in the Singapore Flyer over at Marina the sights of coastal Singapore. Food's plenty along they way so just stop and enjoy anytime. There are free performances in the amphitheatre outside Esplanade for the evening but before that, museums are usually free on Sunday so check the Asian Civilization Museum, Peranakan Museum or the National Museum (they are all within walking distances).
Other than my suggestion, the people here have pretty good advice as well. Depends on what you like really. :-)
