The Singapore Zoo's a definite must! Especially if your kids are interested in learning about animals (what kid isn't!). Universal Studios would be awesome too, what with all the fun rides and more. If you kids are curious about the past you could bring them to the MINT museum (emint.com); they have a collection of retro toys. The Underwater World and Jurong Bird Park might be fun too, depending on if they're interested in marine animals and/or birds.
I wouldn't recommend the Singapore Science Centre; most of the exhibits there are not well-maintained, while Downtown East is more catered to the locals (the Escape Theme Park is a waste of time as it's badly-maintained and there's nothing special about Wild Wild Wet). You'd be much better off at the Singapore Zoo/Sentosa as these are both key tourist attractions.
Shopping-wise, Forum the Shopping Mall is the most children-focused shopping mall on Orchard Road that I know of; there are kids' boutiques, as well as a Toys R Us upstairs.
