We are staying in Venice (Dorsodura) this Sept. and wanting to spend a day exploring the Dolomite Mountains ( Sesto region). Can we catch a train from Venice or should we rent a car for the day? Can anyone highly recommended a specific hiking trail to do?

You can go by train but it is a very very long journey with lots of different changes (at least 8 hours journey each way) www.trenitalia.com. You can go by car, but the journey is still around 3 hours drive if you do not get stuck in traffic. I know there is a bus which leaves from Piazzale Roma for the Dolomites but I am not sure in which place it goes too. That part of the Dolomite is quite far from Venice, there are areas of the Dolomites which are much nearer and just as beautiful. You can take a train or a bus to Cortina for example.
